It occured with all wavelength or only some special wavelength?

If for all, may be the cuvette dirty or the pump is not good.

If not for all, the LED of that wavelength is not good.

Remove flow cell ,now check sensitivity of all wavelenths. or First remove flow cell; go to absorbance mode select 340 nm when the instrument asking for base line press enter

now the instrument will ask for sample , now put back flow cell ( chenk the light path , the flow cell must be in line" with the light path .(LIGHT PATH IS FROM BACK TO FRONT ; NOT RIGHT HAND SIDE TO LEFT HAND SIDE) now press enter

the absorbance must be below 0.3 abs,if not connect peristaltic tubing and give "wash" ; now the absorbance must be below 0.3 abs if not :-

1. either the flow cell is dirty

2. the flow cell not in line with the light path; press the flow cell gently till it sets at the bottom

3. the peristaltic tube is punctured, air bubbles enter making the absorbance too high

I think this is suffiecient to solve the problem now.
