Sectioning

Sectioning thickness is not uniform. sometime sectioning is thick sometime thinner and sometime no sectioning coming out. We are using agarose gel 6% to hold the specimen instead of paraffin.

Are you "double-embedding" - agarose gel first, followed by paraffin? I've seen some discussion online about the methods and techniques for doing that. I've never seen any discussion of sectioning tissue that was embedded only in agarose or similar media; it doesn't seem like that would be firm enough to section properly. However, I am an equipment maintenance specialist, not a histologist.
